How HuSIGHT Works

There are complementary benefits of unbiased autoantibody profiling with peptide libraries versus full-length protein libraries.

Peptide libraries can be very comprehensive in their coverage of all human gene products and their alternatively spliced variants. However, many autoantibodies only recognize the conformational or discontinuous epitopes provided by the 3D folding of full-length proteins.

This type of autoantibody would be undetectable with an assay composed only of peptides. On the other hand, autoantibody detection with an assay composed only of full-length proteins, would be unable to detect the antibodies that target proteins that are difficult to express or simply missing from the library. And in some cases, the importance of an autoantibody is determined not simply by whether the full-length protein is targeted, but rather which specific region or epitope of the protein is targeted.

For these reasons, Infinity Bio has created the first autoantibody profiling assay that combines a comprehensive human peptide library with a large set of full-length human proteins, all in a single, cost-effective assay.

The HuSIGHT-Full_Length sublibrary was constructed from ThermoFisher’s sequence-verified Ultimate ORFeome, which contains 16,516 unique proteins. Request a copy of the HuSIGHT-Full_Length sublibrary.

The HuSIGHT-peptide sublibrary was designed using Infinity Bio’s proprietary approach to multiple sequence alignment-based tiling of all human protein sequences and their variations which are present in the UniProt and RefSeq databases. These overlapping peptide tiles are ~87 amino acid long with ~30 amino acid overlaps, allowing flexibility in length to ensure optimal comparability across homologous regions of a clustered sequence alignment. The optimized human peptidome library design parameters incorporate our decade-plus of experience performing autoantibody profiling with peptide libraries.

Is my human protein of interest present in the library?

Yes, by combining our human full-length and peptide sublibraries, all proteins are represented, with most represented as both peptide tiles and full-length proteins.

How does HuSIGHT compare to phage-based assays?

Phage-display methods are limited to peptide libraries and are therefore unable to detect conformational autoantibodies.

HuSIGHT leverages the unique advantages of MIPSA (Molecular Indexing of Proteins by Self Assembly) to create sublibraries that combine peptide and full-length representation of the human proteome. The VeraCode ™ system for multiplexed barcoding of each antigen provides additional protection from the higher rate of false-positive detection associated with phage display-based methods.

How does HuSIGHT compare to protein arrays?

Protein arrays suffer from the many artifacts associated with the staining and imaging of solid supports as compared to solution-phase assays.

Fluorescence detection methods tend to suffer from a more limited dynamic range compared to DNA sequencing-based technologies.

Full proteome arrays are also typically very costly, often resulting in underpowered studies that report false positive associations or missed subgroup responses. MIPSA provides a cost-effective solution, enabling a unique combination of full-length and peptide sublibraries.
