How It Works

We typically associate ~100 unique barcodes with each library member to optimize both assay robustness and mapping rate.

We use VeraCode™ technology during our library dictionary construction process to:

  • Determine those barcodes associated with unmutated library members
  • Filter your results to these unmutated library members
  • Ensure your data is uncontaminated by mutant antigens and thus entirely interpretable

Three Protein Representation Methods

We provide three different methods for representing sets of proteins using peptides encoded by synthetic oligonucleotides of up to 90 amino acids long.

Simple tiling:

You may select this simple approach to each protein using constant-length peptides of your preferred length and overlap.

Cluster:

You can select to cluster your protein database using multiple sequence alignment (MSA), followed by MSA-based tiling with variable length peptides of your preferred length and overlap.

Proteome sketching:

You can also have us subsample your protein database using epitope prioritization and optional concatenation of shorter peptides to achieve maximal library compression.

Custom MIPSA libraries can also be constructed for deep mutational scanning of important epitopes. Applications include:

  • Fine mapping of antigen-antibody interactions
  • Off-target prediction
  • Analysis of phylogenetic breadth of coverage

Custom Antigen Library FAQs

How long does it take to make a custom library and what is the cost?

Once peptide design is completed, your custom antigen library can be assembled and functionally tested in 4 to 6 weeks. The cost of custom libraries varies due to differences in peptide lengths and overall library complexity.

Is there a size limit for custom libraries?

Our current peptide library size limit is 1,000,000 members. However, please inquire if your library needs exceed this limit, as we may be able to accommodate.

Can I provide my cloned full-length protein coding sequences for incorporation into a MIPSA library?

Most likely. If your previously cloned ORFs are readily compatible with our existing MIPSA library construction, we can incorporate them. Alternatively, we can discuss adaptation or using gene synthesis to recreate them in a compatible format.
